In 2011, a research team led by Professor Shin-Ichiro Imai of the University of Washington School of Medicine (Joint Departments of Developmental Biology and Medicine in St. Louis, Missouri, USA) reported the existence and amazing effects of NMN (also called β-nicotinamide mononucleotide or β-NMN), a precursor of the coenzyme NAD+ (nicotinamide adenine dinucleotide) that belongs to the B3 family of vitamins. This was a significant discovery in the field of medicine.

NMN exists in the cells of all living organisms, and as research on NMN progresses at a global scale, it is attracting more and more interest. NMN is also produced inside the human body naturally, but this production capacity is considered to decrease with age.

As NMN is attracting more and more attention, the market is becoming saturated with NMN products by many companies. However, it is important to realize that the quality of NMN varies from product to product, even in the case of products that claim to contain nothing but NMN.


●Tragedy caused by low purity.

Products containing low-purity NMN obviously contain a great deal of impurities, but what negative effects can be caused by these impurities?

That depends on the impurity. To give an extreme example from the Morinaga case that occurred in Japan in 1955, an additive in milk poisoned more than 10,000 people and killed more than 100. The additive was found to have arsenic in amounts of less than 5%.

Cases like this led government agencies including the United States Food and Drug Administration (FDA) to dramatically increase focus on eliminating impurities.


●NMN purity claims. Absolute and relative measurement methods.

Some products claim to contain NMN with a purity of, for example, 99.9%. At first glance, this may seem very acceptably pure. But what does this 99.9% figure really represent?


・ Purity by the relative measurement method.

Testing institutions compare test samples to standard samples, but in many cases will assume 100% purity of the standard sample.


・ Purity by the absolute measurement method.

There are highly reliable purity test methods that can determine percentages of all  substances detected in an NMN sample. This absolute measurement method is the MIRAILAB method.


As shown in the graph above, taking a "standard NMN" with an absolute purity of 95% and a test NMN from another company with an absolute purity of 94% and carrying out a relative measurement, then raises the purity of the latter to 98.9%.

On the other hand, if we carry out a relative measurement on MIRAILAB NMN in the above scenario, the purity of MIRAILAB NMN rises from 99.8 to 105.0%. The relative measurement method depends on the quality of the "standard NMN" itself, so even if the test result looks good at first glance, that is not necessarily the case. There should be a single high global standard for indicating NMN purity, but at present methods differ and not every company offers a clear explanation.

●Comparative analysis of NMN purity by absolute measurement method

The Institute for Research on Productive Aging (IRPA) recently  analyzed the purity of four NMN samples from as many companies by the absolute measurement method, and found the MIRAILAB sample to be purest.


IRPA carried out two different purity tests (LC-UV (1) and NMR (2)) on the four NMN samples and MIRAILAB NMN was the only sample with purity  of over 99% in both tests.

●Analysis of impurities contained in NMN

The Institute for Research on Productive Aging (IRPA) tested four NMN samples for impurities. The LC-UV method is conducted first, and the separation peaks that can be detected as impurities are tested by mass spectrometry to identify the components of the impurities. The small amount of impurity detected in MIRAILAB NMN was identified as nicotinamide.


Nicotinamide, which is an impurity contained in the NMN of all four companies, is a substance that can be safely ingested by humans as a nutrient, also called niacin or nicotinic acid.

On the other hand, the "OK company" NMN and the "Self-Affirmed GRAS Company" NMN contained not only nicotinamide, but many other impurities as well.

In particular, the NMN of the "Self-Affirmed GRAS Company" contained 6 types of impurities in addition to nicotinamide, and some of them are not considered to be substances found in vivo. Naturally, this raises concerns regarding the kind of adverse effects that can result from the long-term ingestion of NMN with this amount of impurities.


So, even if a product label reads “high-purity NMN,” we cannot always be certain what impurities it may really contain. It should be known that such products are on the market. Purity degree can be low and safety uncertain.

Complete β-NMN and the dangers of enantiomers

What are enantiomers?

Did you know that there are two types of NMN, α--NMN and β-NMN, with different molecular structures?

α-NMN and β-NMN have the same number and type of components (atoms), but the orientation of the connections between these atoms is different, and they are called enantiomers (see Wikipedia).


The differences in the molecular structures of α-NMN and β-NMN are shown schematically below.

α-NMN and β-NMN

Essentially, the NMN we need is β-NMN.

While β -NMN is produced by the human body, unnecessary α-NMN is never produced.

However, the NMN manufactured by simple chemical synthesis for the sake of cost reduction has a high probability of ending up a mixture of α-NMN and β-NMN.

Complete β-NMN, without any α-NMN, is only produced according to an extremely precise manufacturing process.

The costs of such process are therefore high, making complete β-NMN an expensive and valuable substance.


The effects of α-NMN on the human body have not been fully elucidated, as this alpha-form is never produced in the human body as we mentioned above, but we cannot rule out the possibility that α-NMN works in direct opposition to β-NMN in scientific terms. So please be careful, as some anti-aging NMN supplements may be causing the opposite effect.


Based on the selling price of some NMN product currently available on the market, it is highly likely that they are a mixture of β-NMN and α-NMN.


Examples of damage caused by optical isomers

Just like NMN, some medicines that have been used for a long time also have enantiomers.

For example, thalidomide, a component used in sedative and hypnotic drugs, has the enantiomers R-form and S-form. It is now known that R-form thalidomide has sedative and hypnotic properties, while S-form thalidomide is teratogenic, that is, it induces fetal malformations.

At first, however, the effects of the different enantiomers on the human body and organisms were not understood, and the mixture was used as a medicine.

As a result, pregnant women who ingested thalidomide as a mixture of R and S bodies, gave birth to babies with limb malformations: short limbs that resemble those of seals. The thalidomide tragedy, that also includes stillbirths and others, has caused 5800 victims all over the world since 1956, and has brought to the spotlight the dangers of mixing enantiomers.

This incident, that shook the world, was an opportunity to significantly change the pharmaceutical affairs system in Europe, the United States and Japan.
